General description
Alkyne-cholesterol is a clickable cholesterol probe proprietary for Avanti Polar Lipids. They differ from cholesterol by having an alkyne group.
This reagent is a modified lipid containing an omega-terminal alkyne. The terminal alkyne group can be used in a highly specific linking reaction with azide-containing reagents, known as ‘click chemistry′, in the presence of a copper (Cu)-containing catalyst.
Biochem/physiol Actions
Alkyne-cholesterol is not applicable for use in living cells due to copper ion (Cu (I) and Cu (II)) requirements. It aids in easy and sensitive detection of cholesterol using various biophysical methods. Alkyne-cholesterol can be conjugated to boron-dipyrromethene (BODIPY) fluorescent dye. It is useful in cholesterol trafficking studies in various organelles including mitochondria, endoplasmic reticulum and Golgi.

Lipid modifications aid in regulating (and misregulating) protein function and localization. However, efficient methods to screen for a lipid's ability to modify proteins are not readily available. We present a strategy to identify protein-reactive lipids and apply it to a

Cellular cholesterol plays fundamental and diverse roles in many biological processes and affects the pathology of various diseases. Comprehensive and detailed understanding of the cellular functions and characteristics of cholesterol requires visualization of its subcellular distribution, which can be achieved
