General description

JAKs are non-receptor tyrosine kinases which couple to cytokine receptors, and are activated by cytokines. JAK kinases (JAK1, JAK2, JAK3, Tyk2) have a kinase domain and a second, pseudokinase domain which is not enzymatically active. The principal effectors of JAKs are the STATs, which are direct activators of transcription, but JAKs also signal to the Ras-MAP Kinase pathway, and through Tec, to the PI3 Kinase pathway. JAK3 is required for signaling through cytokine receptors which share a common γ chain. JAK2 is required for signaling through a different subset of cytokine receptors, while JAK1 is essential for signaling from the IL-2, IL-6 and interferon receptors. The stress-activated protein kinase 1 (SAPK) family is also referred to as the jun N-terminal kinase family in light of the substrate preference of these serine/threonine kinases. In addition to regulating many biological processes including proliferation and apoptosis, the JNK/SAPK family has been implicated in many neurodegenerative diseases including Alzheimer disease, Parkinson′s disease and Amyotrophic lateral sclerosis.

Recognizes JAK3 (Janus Kinase 3).

Immunogen

Synthetic peptide corresponding to amino acids 782-797 of mouse JAK3 (SDPTPGIPSPRDELC).

Application

Detect JAK3 using this Anti-JAK3 Antibody validated for use in WB & IP.
Immunoprecipitation: 5-10 uL of a previous lot of this antibody was used in immunoprecipitation.

Optimal working dilutions must be determined by end user.

Storage and Stability

Stable for 1 year at -20°C in undiluted aliquots from date of receipt.
Handling Recommendations: Upon first thaw, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
