Biochem/physiol Actions

Protein kinase C-α (PKCα), an isoform of protein kinase C, exhibits an anti-apoptotic and pro-proliferative function. It is commonly upregulated in tumor cells, and its depletion has been associated with apoptosis induction. The anti-apoptotic effects of PKCα are mediated through mechanisms such as phosphorylation of the anti-apoptotic protein B-cell lymphoma 2 (Bcl-2) and activation of the pro-survival protein kinase AKT. The inhibition of PKCα leads to the activation of a PKCδ-dependent apoptotic pathway in salivary epithelial cells. PKCα regulates cell cycle progression in glioma cells by increasing the expression of p21 cyclin-dependent kinase inhibitor 1 (p21 (Waf1/Cip1)), a protein involved in cell cycle regulation.
