General description

The integrin family of cell adhesion receptors consists of at least 16 membrane-associated heterodimers, composed of an alpha and beta subunit that associate in a non-covalent manner. The structure and functional diversity of the integrin family are based upon the pairing abilities of the individual alpha and beta subunits. Key to these molecular interactions between the integrin receptors and their respective ligands is the recognition of the Arg-Gly-Asp (RGD) sequence, known to be present in the extracellular matrix components fibronectin, vitronectin, collagen, fibrinogen, and von Willebrand factor (Cheresh, 1991). The involvement of integrins in vascular proliferation, adhesion, and wound repair has been well documented.

The beta5 subunit associates with alphaV forming a receptor for vitronectin. This complex is expressed in a wide range of cell types including fibroblast and keratinocytes. It is also expressed at high levels in several tumors including lung, pancreatic and mammary carcinomas. It is not expressed in lymphblastoid cells.

Specificity

Recognizes Integrin beta5. Specificity determined by immunoprecipitation from detergent extract of 35S methionine labeled cells. No cross-reactivity to beta1 or beta3.

Immunogen

Epitope: C-terminus/cytosolic
Synthetic peptide derived from the COOH terminal sequence (cytoplasmic domain) of the human integrin beta 5 subunit (KTFNKFNKSYNGTVD). Note : a lysine (K) residue was added to the N-terminal of the peptide to facilitate coupling to the carrier protein.

Application

Detect Integrin β5 using this Anti-Integrin β5 Antibody, C-terminus/cytosolic validated for use in ELISA, IF, IH(P), IP, RIA & WB.
ELISA/RIA

Immunoprecipitation: 5μL of antibody is sufficient to precipitate alphaVbeta5 from

2.5x10E6 cells.

Western blot: non-reduced conditions; do not use milk in the dilution buffer.

Immunofluorescence: 1:100.

Immunohistochemistry (Acetone Fixed) 1:40-1:80

Immunohistochemistry (Paraffin Embedded) 1:200 (ABC)

Optimal working dilutions must be determined by end user.
