General description

Electrogenic sodium bicarbonate cotransporter 1 (UniProt Q9Y6R1; also known as Sodium bicarbonate cotransporter, Na(+)/HCO3(-) co-transporter, Solute carrier family 4 member 4, kNBC1) is encoded by the SLC4A4 (also known as NBC, NBC1, NBCE1) gene (Gene ID 8671) in human. Bicarbonate (HCO3-) transporters are the principal regulators of pH in cells and play a vital role in acid-base movement in various tissues. There exist four types of HCO3- transporters, namely Cl-/-HCO3- exchangers, Na+/HCO3- co-transporters (NBCs), a K+/HCO3- co-transporter, and a Na+-driven Cl-/-HCO3- exchanger. SLC4A4 is a multi-pass membrane NBC with 10 transmembrane segments, 5 extracellular domains, and 6 intracellular domains, having both its N- and C-terminal (a.a. 1-468 and a.a. 987-1079, respectively) exposed intracellularly. NBCs mediate the coupled movement of sodium and bicarbonate ions, in a 3 to 1 stoichiometric ratio, across the plasma membrane. Multiple isoforms are reported as a result of alternative splicing, including NBCe1-A (kNBC1, kNBC-1; UniProt Q9JI66-2/Rat and Q9Y6R1-2/Human) that is mainly expressed in the kidney, NBCe1-B (bNBC1, pNBC-1; UniProt Q9JI66-1/Rat and Q9Y6R1-1/Human) expressed in the pancreas and other tissues, and NBCe1-C (b2NBC; UniProt Q9JI66-3/Rat and Q9Y6R1-5/Human) expressed in the brain.

Specificity

Expected to react with all rat and human spliced isoforms reported by UniProt (Q9JI66 and Q9Y6R1, respectively).

Immunogen

Epitope: Amino acids 338-391
GST-tagged recombinant protein corresponding to amino acids 338-391 of human Na+/HCO3- cotransporter.

Application

Detect Na+/HCO3- Cotransporter using this Anti-Na+/HCO3- Cotransporter Antibody validated for use in Western Blotting, Immunohistochemistry.
Immunohistochemistry Analysis: A 1:1,000 dilution from a representative lot detected Na+/HCO3- cotransporter in human cerebral cortex, kidney and pancreas tissues.
