Specificity

Recognizes a protein of 32 kDa, identified as the Caspase-3 protein. Highly specific to Caspase-3 and shows no cross-reaction with other caspase family members. Caspase-3 is ubiquitously expressed and like other caspases is synthesized as an inactive proenzyme. Upon activation, caspase-3 is cleaved generating two subunits of 17 kDa and 12 kDa respectively. Caspase-3 has been implicated in the induction of apoptosis. Activation of caspase-3 occurs in response to a variety of apoptotic inducers, including Fas-mediated apoptosis.

Western blot: 1 μg/ml for 2 hours at RT.

Immunoprecipitation: 2 μg/mg of protein lysate Immunohistochemistry (frozen and paraffin): 2-4 μg/ml for 30 minutes at RT. Staining of formalin-fixed tissues requires boiling tissue sections in 10 mM citrate buffer, pH 6.0, for 10-20 min. followed by cooling for 20 min. at RT.

Optimal working dilutions must be determined by end user.
