General description

Wnt-1, also known as Proto-oncogene Wnt-1 or Proto-oncogene Int-1 homolog, was first identified as a proto-oncogene activated by viral insertion in mouse mammary tumors. The Wnt-1 gene encodes a member of a large family of secreted proteins that are cysteine-rich, glycosylated, and poorly soluble. Wnt-1 serves as a ligand for the RYK coreceptor, resulting in Wnt signaling. The Wnt signalling pathway mediates the levels of cytosolic beta-catenin, a multi-functional protein that associates with membrane-bound E-cadherin as well as several DNA-binding proteins, such as members of the TCF/LEF family. Additionally, it serves a ligand for members of the frizzled family of seven transmembrane receptors and plays a role in development of the central nervous system.

Purified recombinant fragment of human WNT1 expressed in E. Coli.

Application

Detect Wnt-1 using this mouse monoclonal antibody, Anti-WNT1 Antibody, clone 10C8 validated for use in western blotting, Immunofluorescence, IHC & Flow Cytometry.
Immunofluorescence Analysis: A 1:200-1,000 dilution from a representative lot detected WNT1 in HeLa and 3T3-L1 cells.

Immunohistochemistry Analysis: A 1:200-1,000 dilution from a representative lot detected WNT1 in human adrenal tissue.

Flow Cytometry Analysis: A 1:200-400 dilution from a representative lot detected WNT1 in HeLa cells.

Optimal working dilutions must be determined by end user.

Western Blot Analysis: A 1:500-2,000 dilution of this antibody detected WNT1 in NIH/3T3, 3T3L1, and HeLa cell lysates.

Storage and Stability

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
