General description

Zinc finger protein Aiolos (UniProt: Q9UKT9; also known as Ikaros family zinc finger protein 3, IKZF3) is encoded by the IKZF3 (also known as ZNFN1A3) gene (Gene ID: 22806) in human. IKZF3 is a member of the Ikaros Zinc finger family of transcription factors with six C2H2 type zinc finger domains. It is expressed mainly in peripheral blood leukocytes, the spleen, and the thymus. It is present as a homodimer and can also form heterodimers with other Ikaros family members. It plays an important role in the regulation of lymphocyte differentiation and is involved in the regulation of B-cell differentiation, proliferation and maturation. It is also shown to regulate Bcl-2 expression and controls apoptosis in T-cells in an IL2-dependent manner. Its levels are shown to be upregulated by TGF-1. IKZF3 expression Th17 cells is associated with increased IL-10 production and it is shown to interact with known regulators of IL10 expression. It also acts as a transcriptional activator in CD4+ T cells in cooperation with other factors, such as FOXP3 and BLIMP1 in CD4+ regulatory T cells (Tregs). The degron sequence of IKZF3 is localized to amino acids 144-168 and a Q147H mutation is reported to cripple the function of IKZF3. This sequence constitutes a modular degron that can be used to target heterologous proteins. This degron sequence is as capable as the full-length IKZF3 at targeting selected proteins for degradation. It is shown to target green fluorescent protein (GFP) for pomalidomide-induced degradation when fused to the C-terminus of GFP, but not to the N-terminus. It has also been sown to target exogenous HIF2 for pomalidomide-induced degradation when fused to the C-terminus of HIF2 . Clone 1C3-D3-F8 recognizes the degron sequence that is considered as an important regulator of protein degradation. (Ref.: Koduri, V. et al. (2019). Proc. Natl. Acad. Sci. USA. 116(7); 2539-2544).

Specificity

Clone 1C3-D3-F8 is a mouse monoclonal antibody that detects IKZF3 Protein. It targets an epitope within 25 amino acids from a Degron sequence in the N-terminal half.

KLH-conjugated linear peptide corresponding to Degron sequence from human IKZF3.

Application

Quality Control Testing

Evaluated by Western Blotting in lysate from HEK293 cells expressing IKZF3 Degron-tagged PTPA.

Western Blotting Analysis (WB): A 1:500 dilution of this antibody detected Zinc finger protein Aiolos (IKZF3) in lysates from HEK293 cells ectopically expressing IKZF3 Degron-tagged PTPA, but not in lysate from untransfected HEK293 cells (wild type).

Storage and Stability

Recommend storage at +2°C to +8°C. For long term storage antibodies can be kept at -20°C. Avoid repeated freeze-thaws.
