Clone 3758-A37-5 is a mouse monoclonal antibody that detects Rho-type GTPase-activating protein 11B (ARHGAP11B). It targets an epitope within 21 amino acids from the C-terminal region.

KLH-conjugated linear peptide corresponding to 21 amino acids from the C-terminal region of human Rho-type GTPase-activating protein 11B (ARHGAP11B).

Application

Quality Control Testing

Evaluated by Western Blotting in lysate from COS-7 cells transfected with human ARHGAP11B.

Western Blotting Analysis: A 1:500 dilution of this antibody detected ARHGAP11B in lysate from COS-7 cells transfected with human ARHGAP11B, but not in lysate from wild-type COS-7 cells.

Tested Applications

Target description

Inactive Rho GTPase-activating protein 11B (UniProt: Q3KRB8; also known as Rho-type GTPase-activating protein 11B) is encoded by the ARHGAP11B (also known as FAM7B1) gene (Gene ID: 89839) in human. ARHGAP11B is a mitochondrial matrix protein that is specifically expressed in radial glia cells of cortical neural stem and progenitor cells during neocortex development. It is preferentially expressed in apical and basal radial glia and is not detected in cortical neurons and cortical plate. It promotes amplification of basal progenitors in the subventricular zone, producing more neurons during fetal corticogenesis, thereby playing a key role in neocortex expansion. It promotes basal progenitor cell proliferation by inhibiting the mitochondrial permeability transition pore (mPTP). It delays the opening of the mPTP via interaction with ADP:ATP translocase, thereby increasing mitochondrial Ca2+ concentration and inducing glutamine catabolism, which is required for basal progenitor proliferation. ARHGAP11B does not exhibit GTPase activator activity and this lack of GTPase activator activity is essential for amplification of basal progenitors during neocortex development.
