General description

E3 ubiquitin-protein ligase MYCBP2 (UniProt: O75592; also known as Myc-binding protein 2, Pam/highwire/rpm-1 protein, Protein associated with Myc, MYCBP2) is encoded by the MYCBP2 (also known as KIAA0916, PAM) gene (Gene ID: 23077) in human.MYCBP2 protein mediates the ubiquitination and subsequent proteasomal degradation of TSC2/tuberin. It is ubiquitously expressed, but brain and thymus display exceptionally abundant expression. It is shown to co-localize with TSC1 and TSC2 along the neurites and in the growth cones. MYCBP2 may also function as a facilitator or regulator of transcriptional activation by MYC and have a role during synaptogenesis. It is reported to negatively regulate neuronal growth, synaptogenesis, and synaptic strength. Mice with MYCBP2-deficiency in peripheral sensory neurons show prolonged thermal hyperalgesia. Loss of MYCBP2 is also linked to constitutively activated p38 MAPK and increased expression of several proteins involved in receptor trafficking. Down-regulation of MYCBP2 in the spinal cord of adult rats is shown to enhance their. MYCBP2 is also shown to be an inhibitor of adenylyl cyclase. (Ref.: Holland, S et al. (2011). J. Biol. Specificity

Clone PM-8A9 specifically detects MYCBP2 in HEK293 cells. It targets an epitope within the first 150 amino acids in the N-terminal region.
