General description

Ankyrin repeat and sterile alpha motif domain-containing protein 1B (UniProt: P0C6S7; also known as Amyloid-beta protein intracellular domain-associated protein 1, AIDA-1, E2A-PBX1-associated protein, EB-1) is encoded by the Anks1b gene (Gene ID: 314721) in rat. AIDA-1 is an adaptor protein that has been implicated in Cajal body and nucleolar function. It is expressed throughout the brain and is highly expressed in the hippocampus. It is one of the most abundant proteins at postsynaptic densities where it is reported to bind to PSD95 and associates with NMDA receptors (NMDARs). In the brain it interacts with amyloid b precursor protein and plays a role in brain development. AIDA-1 preferentially associates with GluN2B-containing NMDARs and plays a role in their transport from the endoplasmic reticulum (ER). Loss of AIDA-1 results in the accumulation of GluN2B in the ER and decreases GluN2B and increases GluN2A distribution at the synapse. However, loss of AIDA-1 does not affect AMPAR-mediated synaptic transmission or the probability of glutamate release. Mutations in Anks1b gene are known to affect GluN2B function and are linked to the pathogenesis of neuropsychiatric disorders. (Ref.: Carbonell, AU., et al., (2019). Nat. Commun. 10(1); 3529; Tindi, JO., et al. (2015). J. Neurosci. 35(24); 8986- 8996).

Specificity

Clone 1A11 is a mouse monoclonal antibody that detects AIDA-1 in multiple mammalian species. It targets an epitope within 10 amino acids from the SAM2 domain in the C-terminal half.

Immunogen

A linear peptide corresponding to 10 amino acids from the SAM2 domain within the C-terminal half of rat Ankyrin repeat and sterile alpha motif domain-containing protein 1B (AIDA-1).

Application

Quality Control Testing

Evaluated by Western Blotting in Rat brain tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected AIDA1 in Rat brain tissue lysate.

Tested Applications

Western Blotting Analysis: A representative lot detected AIDA1 in Western Blotting applications (Tindi, J.O., et al. (2015). J Neurosci. 35(24): 8986-96).

Immunofluorescence Analysis: A representative lot detected AIDA1 in Immunofluorescence applications (Tindi, J.O., et al. (2015). J Neurosci. 35(24): 8986-96).

Immunohistochemistry (Paraffin) Analysis (IHC(P)): A 1:50 dilution from a representative lot detected AIDA1 in rat cerebral cortex, hippocampus, and cerebellum tissue sections.

Immunoprecipitation Analysis: A representative lot immunoprecipitated AIDA1 in Immunoprecipitation applications (Carbonell, A.U., et al. (2019). Nat Commun. 10(1): 3529).

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user
Anti-AIDA1, clone 1A11, Cat. No. MABN2639, is a mouse monoclonal antibody that detects AIDA1/ANKS1B and is tested for use in Immunofluorescence, Immunohistochemistry (Paraffin), Immunoprecipitation, and Western Blotting.

Physical form

Purified mouse monoclonal antibody IgG2b in buffer containing 0.1 M Tris-Glycine (pH 7.4), 150 mM NaCl with 0.05% sodium azide.
