General description

Human SYNE1 gene (UniProt Q8NF91; Gene ID 23345) is among the top ten loci conferring risk for the five major psychiatric disorders, schizophrenia, bipolar disorder (BD), major depression, attention deficit-hyperactivity disorder (ADHD), and autism spectrum disorder (ASD), as well as multiple other neurological and neuromuscular disorders. The BD-associated single nucleotide polymorphisms (SNPs) are in a region of the human SYNE1 gene homologous to brain-specific transcripts of rat Syne1-encoding CPG2 (candidate plasticity gene 2; UniProt Q63128; Gene ID 499010), originally identified as an upregulated transcript during kainic acid-induced seizures in the rat dentate gyrus. The rat CPG2 spliced variant localizes to excitatory synapses of excitatory neurons, where it plays a role in synaptic plasticity and facilitates glutamate receptor cycling.

Specificity

Immunogen sequence is present in rat spliced isoform CPG2 (UniProt Q63128), as well as in mouse spliced isoform Enaptin (a.k.a. Nesprin-1 Giant), Syne-1B, and Enaptin-165 (UniProt Q6ZWR6). Target region is not found in rat isoform Nesprin-1 (UniProt Q8VHJ9), or in mouse isoform 2 and Syne-1A. Clone 200A6 detected shRNA-mediated knockdown of SYNE1 spliced isoform CPG2 in rat neurons by fluorescent immunocytochemistry and immunoblotting (Cottrell, J.R., et al. (2004). Neuron. 44(4):677-690).
This clone also detects SYNE1 protein in human brain samples with a much weaker signal than that observed in rat samples.

Immunogen

Synthetic peptide corresponding to a sequence within the third coiled coil of rat spliced isoform CPG2 or the tenth Spectrin domain of mouse spliced isoform 1 (a.k.a. Enaptin or Nesprin-1 Giant).

Application

Anti-SYNE1, clone 200A6, Cat. No. MABN857, is a highly specific mouse monoclonal antibody that targets SYNE1 splice variants, including rat CPG2, and has been tested in Electron Microscopy, Immunocytochemistry, and Western Blotting.
Electron Microscopy Analysis: A representative lot localized SYNE1 isoform CPG2 immunoreactivity concentrated in the postsynaptic site of excitatory synapses, in a distinct synaptic subdomain lateral to and beneath the postsynaptic density (PSD) by immunoEM labeling of 4% paraformaldehyde-/0.1% glutaraldehyde-fixed, lowicryl-embedded rat hippocampal cryosections (Cottrell, J.R., et al. (2004). Neuron. 44(4):677-690).

Electron Microscopy Analysis: A representative lot localized SYNE1 isoform CPG2 immunoreactivity colocalized with clathrin in the postsynaptic endocytic zone by immunoEM labeling of 60 nm thick serial sections of 4% paraformaldehyde-/0.1% glutaraldehyde-fixed rat neurons (Cottrell, J.R., et al. (2004). Neuron. 44(4):677-690).

Immunocytochemistry Analysis: A representative lot immunolocalized SYNE1 spliced isoform CPG2 on the dendritic spines, colocalized with clathrin, but segregated from the PSD and actin filaments in the spine head, by fluorescent immunocytochemistry staining of 4% formaldehyde-fixed, 0.3% Triton X-100-permeabilized rat hippocampal neurons. Target staining was greatly diminished among neurons received cellular CPG2 shRNA delivery (Cottrell, J.R., et al. (2004). Neuron. 44(4):677-690).

Western Blotting Analysis: A representative lot detected SYNE1 spliced isoform CPG2 as a ~110 kDa boublet band in rat cerebral cortex tissue extract and lysate from isolated rat cortical neurons. Target band detection was greatly diminished when using lysate from neurons following cellular CPG2 shRNA delivery (Cottrell, J.R., et al. (2004). Neuron. 44(4):677-690).

Quality

Evaluated by Western Blotting in rat hippocampus tissue lysate.

Western Blotting Analysis: 4 µg/mL of this antibody detected the ~110 kDa SYNE1 spliced isoform CPG2 in 10 µg of rat hippocampus tissue lysate.

Target description

~110 kDa (rat isoform CPG2) observed. 109.0 kDa (rat isoform CPG2), 1,010 kDa (mouse isoforms Giant/Enaptin and Syne-1B), and 165.2 kDa (mouse isoform Enaptin-165) calculated. Uncharacterized bands may be observed in some lysate(s).
