General description

Kinases, alternatively referred to as phosphotransferases, form an extensive family of enzymes responsible for transferring phosphate groups from the high-energy donor molecule ATP to their designated substrates. This family of enzymes is renowned for its role in regulating a myriad of cellular processes, with the protein kinases constituting the largest subgroup. In humans, 518 distinct kinases have been identified, and a substantial portion of human proteins, up to 30%, undergo modification by these kinases. The considerable diversity and pivotal role of kinases in cellular signaling position them as promising targets for drug development, biochemical and biomedical research.

Application

The Kinase Assay Kit may be used for:
  • Cancer Research
  • Cell Signaling Research
  • Drug Discovery

Features and Benefits

  • High Sensitivity and Accuracy: Achieve quantification of kinase activity, detecting as low as 0.01 U/L with high sensitivity and accuracy.
  • Streamlined Procedure: Effortlessly conduct the assay with a simplified process, adding just a single working reagent. Complete the assay in a quick 10-minute reaction, optimizing efficiency and saving valuable time.
  • High-Throughput Inhibitor Screening: Screen for kinase inhibitors effectively with a minimal 5 μL test compound requirement, enhancing efficiency in high-throughput settings.

Suitability

The kit is suitable for the determination of protein kinase activity and for high-throughput screening of kinase inhibitors.

Principle

The Kinase Assay Kit provides a simple and rapid method for assaying kinase activity and high-throughput screening for kinase inhibitors. This homogeneous microplate-based assay involves incubating the kinase with a single working reagent, in which ADP is enzymatically converted to ATP and pyruvate, which is quantified using a fluorometric (Ex = λ530 nm/λEm = 590 nm) assay method.
