General description

Ras-related protein Rab-14 (Rab14) is a member of the Rab11 subfamily of small guanosine triphosphatases (GTPases). It is associated with the early endosomal clathrin-coated trans‐Golgi network microdomain and is ubiquitously expressed. Rab14 belongs to the RAS oncogene family and is mapped to human chromosome 9q33.2.

Specificity

Anti-Rab14 recognizes human, rat, and mouse Rab14 (not tested in other species).

Application

Anti-Rab14 antibody produced in rabbit has been used in western blotting and immunofluorescence.

Biochem/physiol Actions

Ras-related protein Rab-14 (Rab14) is associated with the trafficking of the fibroblast growth factor receptor 2 (FGFR2) and regulates N-cadherin turnover. It is involved in membrane trafficking between the Golgi complex and endosomes. In polarized epithelial cells, Rab 14 regulates delivery of cargo from the trans‐Golgi network (TGN) to the apical membrane domain. Rab14 has also been implicated in phagosomal biogenesis. It acts as a tumor oncogene in non-small cell lung carcinoma (NSCLC) and is upregulated in gastric cancer.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2–8 °C for up to one month. For extended storage, freeze in working aliquots. Repeated freezing and thawing, or storage in “frost-free” freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
