General description

Pyruvate dehydrogenase kinase isoenzyme 4 (PDK4) is a mammalian mitochondrial serine kinase, encoded by the gene mapped to human chromosome 7q21.3. The encoded protein belongs to the PDK family. PDK4 transcripts are highly expressed in heart and skeletal muscle. It is also expressed at moderate levels in human brain, placenta, lung, liver, kidney and pancreas.
This gene is a member of the PDK/BCKDK protein kinase family and encodes a mitochondrial protein with a histidine kinase domain. This protein is located in the matrix of the mitrochondria and inhibits the pyruvate dehydrogenase complex by phosphorylating one of its subunits, thereby contributing to the regulation of glucose metabolism. Expression of this gene is regulated by glucocorticoids, retinoic acid and insulin. (provided by RefSeq)

Biochem/physiol Actions

Pyruvate dehydrogenase kinase isoenzyme 4 (PDK4) catalyzes the phosphorylation of pyruvate dehydrogenase (PDH and also facilitates ATP-dependent inactivation of the PDH complex. Thus, it plays a vital role in glucose utilization and lipid metabolism. Elevated expressions of the protein leads to progressive small vessel loss in pulmonary arterial hypertension (PAH) and reduced endothelial-pericyte interactions. PDK4 is considered to be an emerging therapeutic target for type 2 diabetes.
