General description

Monoclonal Anti-USP8 (mouse IgG1 isotype) is derived from the hybridoma US872 produced by the fusion of mouse myeloma cells and splenocytes from BALB/c mice. Ubiquitin specific protease 8 (USP8) belongs to the family of de-ubiquitinating enzymes (DUBs).

Immunogen

synthetic peptide corresponding to an internal sequence of human USP8, conjugated to KLH.

Application

Monoclonal Anti-USP8 antibody produced in mouse has been used in:immunoblotting immunofluorescence,flow cytometry, immunocytochemistry.

Biochem/physiol Actions

Ubiquitin specific protease 8 (USP8) was found to regulate ubiquitination of frizzled, a receptor for Wnt/Wingless. Deletion of USP8 in mice causes embryonic lethality. The defect was accompanied by a strong reduction or absence of several growth factor receptor tyrosine kinases (RTKs), like EGFR, hepatocyte growth factor receptor (c-met), and ERBB3.USP8 inactivation results in enhanced ubiquitination of ligand-activated EGFR. USP8 in conjunction with components of the ESCRT-0 complex, plays an integral role in the early endosomal sorting machinery that functions to protect epidermal growth factor receptor (EGFR) as well as other receptors from lysosomal degradation.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
