General description

Human Jak1 (Janus kinase 1), also known as JTK3, (GenBank Accession No. NM_002227), amino acids 866-1155 (end) with N-terminal GST-Tag, MW= 59 kDa, expressed in Sf9 cells via a Baculovirus expression system.
JAK (Janus kinases) are activated when cytokines bind with their receptors, upon which they phosphorylate the receptors and produce docking sites for signaling molecules. One of the main signaling molecules includes signal transducer and activator of transcription (Stat) family. JAK1 is needed by multiple cytokines, including the family using the shared receptor subunit called common γ chain (γc), such as interleukin (IL)-2, IL-4, IL-7, IL-9, IL-15 and IL-21. JAK1 KO (knockout) mice have early postnatal lethal or perinatal lethal phenotype due to neurological defects. In vitro studies show that JAK1S703I mutation functions as an activator of JAK-STAT pathways even in the absence of cytokines, and drives cellular proliferation. JAK1S703I mutant patient-derived xenograft (PDX) HCC (hepatocellular carcinoma) cells also show sensitivity to JAK1/2 inhibitor, ruxolitinib.

Application

Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling.

Formulated in 40 mM Tris-HCl pH 8.0, 110 mM NaCl, 2.2 mM KCl, 3 mM DTT,16 mM glutathione and 20% glycerol.
