Specificity

Clone 1D5 is a ZooMAb rabbit recombinant monoclonal antibody that specifically detects human Secretory phospholipase A2 receptor (PLA2R1). It targets an epitope with in the extracellular domain.

Immunogen

GST/His-tagged recombinant fragment corresponding to 136 amino acids from the N-terminal, extracellular domain region of human Secretory phospholipase A2 receptor (PLA2R1).

Application

Anti-PLA2R1, clone 1D5, ZooMAb, Cat. No. ZRB012657, is a recombinant Rabbit Monoclonal antibody that detects Secretory phospholipase A2 receptor and has been tested for use in Immunocytochemistry, Immunohistochemistry (Paraffin), and Western Blotting.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected PLA2R1 in HeLa, A431, HUVEC and NIH 3T3 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected PLA2R1 in human kidney tissue sections.

Target description

Secretory phospholipase A2 receptor (UniProt: also known as PLA2-R, PLA2R, 180 kDa secretory phospholipase A2 receptor, C-type lectin domain family 13-member C, M-type receptor) is encoded by the PLA2R1 (also known as CLEC13C) gene (Gene ID: 22925) in human. PLA2R is a single-pass type I membrane protein that serves as a receptor for secretory phospholipase A2 (sPLA2). It is highly expressed in kidney and is also present in lung macrophages, pancreas, amnion, and placenta. PLA2R is a receptor for phospholipase sPLA2-IB/PLA2G1B, but not sPLA2-IIA/PLA2G2A. It can also bind to snake PA2-like toxins. Binding of sPLA2 to its receptor participates in both positive and negative regulation of sPLA2 functions and clearance. Binding of sPLA2-IB/PLA2G1B is shown to induce various effects depending on the cell type, such as activation of the mitogen-activated protein kinase cascade to induce cell proliferation, the production of lipid mediators, selective release of arachidonic acid in bone marrow-derived mast cells. PLA2R is also involved in responses in proinflammatory cytokine productions during endotoxic shock. The circulating, soluble form of secretory phospholipase A2 receptor acts as a negative regulator of sPLA2 functions by blocking the biological functions of sPLA2-IB/PLA2G1B. PLA2R is initially produced with a signal peptide sequence (aa 1-20), the cleavage of which produced the mature form that has a large extracellular domain (aa 21-1397), a transmembrane domain (aa 1398-1418), and a cytoplasmic tail (aa 1419-1463). The extracellular domain contains a fibronectin-like type II domain (173-221), and a tandem repeat of eight carbohydrate-recognition domains (C-type lectins). This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

120 μg/mL after reconstitution at 25 μL. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
