Specificity

Clone 5J13 ZooMAb is a rabbit recombinant monoclonal antibody that specifically detects TATA-box-binding protein. It targets an epitope with in 57 amino acids from the N-terminal region.

Immunogen

GST/Hig-tagged recombinant fragment corresponding to the first 57 amino acids from the N-terminal region of human TATA-box-binding protein.

Application

Anti-TATA-Binding-Protein, clone 5J13 ZooMAb , Cat. No. ZRB1106, is a recombinant rabbit monoclonal antibody that specifically targets TATA-Binding-Protein and has been tested for use in Immunocytochemistry, Immunohistochemistry (Paraffin), and Western Blotting.
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected TATA-Binding-Protein in rat testis and HeLa cell lysate.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected TATA-Binding-Protein in HeLa, A431, HUVEC and NIH 3T3 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected TATA-Binding-Protein in human colon and mouse colon tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

TATA-box-binding protein (UniProt: P20226; also known as TATA sequence-binding protein, TATA-binding factor, TATA-box factor, Transcription initiation factor TFIID TBP subunit) is encoded by the TBP (also known as GTF2D1, TF2D, TFIID) gene (Gene ID: 6908) in human. TATA-box-binding protein is a general transcription factor that functions at the core of the DNA-binding multiprotein factor TFIID. Binding of TFIID to the TATA box is the initial transcriptional step of the pre-initiation complex (PIC), playing a role in the activation of eukaryotic genes transcribed by RNA polymerase II. It is a part of the RNA polymerase II eukaryotic transcription complex and is required for the transcriptional initiation of RNA polymerases. TATA-box-binding protein is widely expressed, with levels highest found in the testis and ovary. Mutations in TBP result in spinocerebellar ataxia type 17(SCA17), a cerebellar disorder classified as autosomal dominant cerebellar ataxia and characterized by atrophy to the cerebellar and cerebral regions, extrapyramidal signs, and dementia. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose. Normal appearance is a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

30 μg/mL after reconstitution at 25 μL. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
