Clone 1J23 ZooMAb is a rabbit recombinant monoclonal antibody that specifically detects human Recoverin.

Application

Anti-Recoverin, clone 1J23 ZooMAb , Cat. No. ZRB1107, is a recombinant rabbit monoclonal antibody that specifically targets Recoverin and has been tested for use in Immunocytochemistry, Immunofluorescence, Immunohistochemistry (Paraffin), and Western Blotting.
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Recoverin in ARPE-19 cells treated with 3 uM fenretinide for 7 days.

Immunofluorescence Analysis: A 1:1,250 dilution from a representative lot detected Recoverin in human retina tissue sections.

Immunohistochemistry (Paraffin) Analysis: A 1:1,250 dilution from a representative lot detected Recoverin in human retina tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Recoverin (UniProt: P35243; also known as Cancer-associated retinopathy protein, Protein CAR) is encoded by the RCVRN (also known s RCV1) gene (Gene ID: 5957) in human. Recoverin is a member of the EF-hand neuronal calcium sensor (NCS) proteins that is expressed in retinal photoreceptors and midget cone bipolar cells. It unctions as a calcium sensor that regulates rhodopsin phosphorylation through inhibition of rhodopsin kinase. It contains four EF hand motifs (aa 25-60; 61-96; 97-132; 147-182) and a myristoyl chain on the N-terminus. It has a high affinity (aa 110-121) and a low affinity (aa 74-85) calcium binding site. Recoverin undergoes a conformational change upon calcium binding in darkness and subsequently it attaches to the disc membrane in photoreceptor outer segment. This in turn allows binding of G protein-coupled receptor kinases (GRK). Calcium-bound Recoverin is shown to inhibit G-protein-coupled receptor kinase (GRKs) in darkness, which prevents visual pigment phosphorylation. Under bright light condition, calcium dissociates from Recoverin and the protein translocates to the inner segment. The released GRK can phosphorylate and deactivate the light activated visual pigment. Mutations in Recoverin have been linked to retinal dysfunction associated with early-stage diabetes and cancer-associated retinopathy. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose. Normal appearance is a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

30 μg/mL after reconstitution at 25 μL. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Stable for 1 year at 2-8°C from date of receipt.Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
