Specificity
Clone 5C20 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Zinc finger E-box-binding homeobox 1 (ZEB1). It targets an epitope within 22 amino acids from the C-terminal half. It is expected to react with all isoforms.
Immunogen
KLH-conjugated linear peptide corresponding to 22 amino acids from the C-terminal half of human Zinc finger E-box-binding homeobox 1 (ZEB1).
Application
Quality Control Testing
Evaluated by Western Blotting in Jurkat cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected ZEB1 in Jurkat cell lysate.
Tested applications
ELISA Analysis: 300 μg/mL from a representative lot detected ZEB1 in ZEB1 in an ELISA.
Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected ZEB1 in HeLa, NIH3T3, HEPG2, A431, and Jurkat cells.
Affinity Binding Assay: A representative lot of this antibody bound ZEB1 with a KD of 2.8 x 10-6 in an affinity binding assay.

Target description
Zinc finger E-box-binding homeobox 1 (UniProt: P37275; also known as NIL-2-A zinc finger protein, Negative regulator of IL2, Transcription factor 8, TCF-8) is encoded by the ZEB1 (also known as AREB6, TCF8) gene (Gene ID: 6935) in human. ZEB1 is a transcription factor that is characterized by the presence of two C2H2-type flanking zinc finger clusters, which are responsible for interaction with paired CACCT(G) E-box-like promoter elements on DNA, and a centrally located POU-like homeodomain, not binding DNA. It is a prime element of a network of transcription factors controlling EMT and has been identified as an important molecule in the regulation of DNA damage, cancer cell differentiation, and metastasis. ZEB1 expression is associated with aggressive behavior, metastasis, treatment resistance and poor prognosis in different tumor types, including breast, pancreatic and lung cancer. In breast cancer, highest ZEB1 expression is observed in the aggressive triple negative and basal types. ZEB1 acts as a transcriptional repressor and inhibits interleukin-2 gene expression. It can enhance or repress the promoter activity of the ATP1A1 gene depending on the quantity of cDNA and on the cell type. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.(Ref.: Drapela, S., et al., (2020). Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb® formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
