Specificity
Clone 1O6 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects NAD-dependent protein deacetylase sirtuin-6 (SIRT6). It targets an epitope within 15 amino acids from the N-terminal region.
Immunogen
KLH-conjugated linear peptide corresponding to 15 amino acids from the N-terminal region of human NAD-dependent protein deacetylase sirtuin-6 (SIRT6).
Application
Quality Control Testing
Evaluated by Western Blotting in Rat Brain tissue lysate.
Western Blotting Analysis: A 1:10,000 dilution of this antibody detected SIRT6 in Rat Brain tissue lysate.
Tested applications
Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected SIRT6 in A431 and NIH3T3 cells.
Affinity Binding Assay: A representative lot of this antibody bound SIRT6 peptide with a KD of 1.0 x 10-12 in an affinity binding assay.

Target description
NAD-dependent protein deacetylase sirtuin-6 (UniProt: Q8N6T7; also known as EC:2.3.1.286, Regulatory protein SIR2 homolog 6, SIR2-like protein 6, SIRT6) is encoded by the SIRT6 (also known as SIR2L6) gene (Gene ID: 51548) in human. SIRT6 is a zinc-dependent nuclear enzyme that is involved in various cellular processes, including telomere maintenance, gene expression, genomic stability, cell senescence, and apoptosis. It is associated with telomeric heterochromatin regions in the nucleus. In mitosis, endogenous SIRT6 is shown to localize to the cytoplasm and partially co-localizes with mitotic spindles, and does not associate with condensed chromatin. It is highly expressed in skeletal muscle, heart, liver, and brain where it affects transcriptional regulation in a tissue-specific manner. SIRT6 promotes resistance to DNA damage and oxidative stress, the two principal defects associated with age-related diseases. It has a two-domain structure consisting of a large Rossmann fold and a smaller and structurally varied region that contains the Zinc-binding motif. Its C-terminus is required for its proper nuclear localization, while the N-terminus is important for chromatin association and for its intrinsic catalytic activity. It displays very weak deacetylase activity and can bind NAD+ in the absence of acetylated substrate. It is shown to modulates acetylation of histone H3 in telomeric chromatin during the S-phase of the cell cycle. SIRT6 deficiency results in genomic instability and hypersensitivity to ionizing radiation. Cells deficient in SIRT6 exhibit higher levels of HIF1 activity, increased glucose uptake, and reduced mitochondrial respiration. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
