Specificity

Clone 1F4 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Mitochondrial Rho GTPase 1 (MIRO-1). It targets an epitope within 22 amino acids from the N-terminal, cytoplasmic domain.

Immunogen

KLH-conjugated linear peptide corresponding to 22 amino acids from the N-terminal, cytoplasmic domain of human Mitochondrial Rho GTPase 1 (MIRO-1).

Application

Quality Control Testing

Evaluated by Western Blotting in A549 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected RHOT1/MIRO-1 in A549 cell lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected RHOT1/MIRO-1 in human skeletal muscle and NIH3T3 cell lysates.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected RHOT1/MIRO-1 in A549 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected RHOT1/MIRO-1 in human heart tissue sections.

Target description

Mitochondrial Rho GTPase 1 (UniProt: Q8IXI2; also known as MIRO-1, hMiro-1, Rac-GTP-binding protein-like protein, Ras homolog gene family member T1) is encoded by the RHOT1 (also known as ARTH1) gene (Gene ID: 55288) in human. MIRO-1 is a single-pass type IV membrane protein found on the outer mitochondrial membrane. It contains a large cytoplasmic domain (aa 1-592), a transmembrane domain (aa 593-615), and a short mitochondrial intermembrane domain (aa 616-618). It contains structurally distinct N-terminal and C-terminal GTP-binding motif with a linker region (MiroS) connecting two EF-hand domains (aa 184-219 and 304-339) to the C-terminal GTPase domain. It also contains two ligand-mimicking -helices (LM1 and LM2) that connect each canonical EF hand to a non-canonical hidden EF-hand domain. The calcium-binding amino acids (aa 197-208 and 317-328) of MIRO-1 are exposed to the cytosol via a helix-loop-helix-motif in both EF hands, facilitating a conformational change of the protein upon binding to calcium. MIRO-1 is involved in mitochondrial trafficking and control of anterograde transport of mitochondria and their subcellular distribution. It undergoes ubiquitination by parkin that leads to its degradation and enhancement of mitophagy. Proteins encoded by several Parkinson s disease linked genes are reported to physically interact with MIRO-1 and modify its function and contribute to the dysregulation of neuronal integrity. Neuron-specific loss of MIRO-1 in a murine model is shown to cause depletion of mitochondria from corticospinal tract axons and progressive neurological deficits that mirrors human upper motor neuron disease. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
