Specificity

Clone 1B2 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Multidrug resistance-associated protein 2 ( MRP2). It targets an epitope within 14 amino acids from the C-terminal, cytoplasmic domain.

Immunogen

KLH-conjugated linear peptide corresponding to 14 amino acids from the C-terminal, cytoplasmic domain of human Multidrug resistance-associated protein 2 ( MRP2).

Application

Quality Control Testing

Evaluated by Western Blotting in A549 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected MRP2 in A549 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected MRP2 in Human liver tissue lysate.

Flow Cytometry Analysis: 0.1 μg from a representative lot detected MRP2 in one million A549 cells.

Affinity Binding Assay: A representative lot of this antibody bound MRP2 peptide with a KD of 1.3 x 10-7 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected MRP2 in A549 cells.

Immunohistochemistry (Paraffin) Analysis: A 1:500 dilution from a representative lot detected MRP2 in human liver tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

ATP-binding cassette sub-family C member 2 (UniProt: Q92887; also known as Canalicular multidrug resistance protein, Canalicular multi-specific organic anion transporter 1, Multidrug resistance-associated protein 2, MRP2) is encoded by the ABCC2 (also known as CMOAT, CMOAT1, CMRP, MRP2) gene (Gene ID: 1244) in human. MRP2, a member of the family of human ATP-binding cassette (ABC) transporters, binds and hydrolyzes ATP to enable active transport of various substrates, including many drugs, toxicants, and endogenous compound across cell membranes. It transports a wide variety of conjugated organic anions such as sulfate-, glucuronide- and glutathione (GSH)-conjugates of endo- and xenobiotics substrates. It is reported to mediate hepatobiliary excretion of mono- and bis-glucuronidated bilirubin molecules. Hence, it plays an important role in bilirubin detoxification. MRP2 is expressed in the apical hepatocyte plasma membrane, renal proximal tubules, and small intestine, where it plays a role in the elimination and oral bioavailability of drugs, xenotoxins, and their metabolites. It also confers resistance to several anti-cancer drugs, including cisplatin, doxorubicin, epirubicin, methotrexate, etoposide, and vincristine. It contains two conserved nucleotide-binding domains (aa 671-678 and 1334-1341) that mediate the binding and hydrolyzing ATP via an ATPase, thereby providing energy for substrates export from the cytoplasm to the extracellular space. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

30 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
