Specificity

Clone 1D21 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Myocyte enhancer factor 2C (MEF2C). It targets an epitope within 17 amino acids from the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 17 amino acids from the N-terminal half of human Myocyte enhancer factor 2C (MEF2C).

Application

Quality Control Testing

Evaluated by Western Blotting with His-tagged recombinant fragment of Human MEF2C.

Western Blotting Analysis (WB): A 1:1,000 dilution of this antibody detected His-tagged recombinant fragment of Human MEF2C.

Tested Applications

Immunohistochemistry (Paraffin) Analysis: A 1:100-1,000 dilution from a representative lot detected MEF2C in human cerebral cortex and human tonsil tissue sections.

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected MEF2C in HeLa cells.

Affinity Binding Assay:: A representative lot of this antibody bound MEF2C peptide with a KD of 1.7 x 10-7 in an Affinity Binding Assay.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Myocyte-specific enhancer factor 2C (UniProt: Q06413; also known as Myocyte enhancer factor 2C) is encoded by the MEF2C gene (Gene ID: 4208) in human. Myocyte enhancer factor 2 (MEF2) family of transcription factors in vertebrates consists of four members: MEF2A, MEF2B, MEF2C, and MEF2D. They are highly conserved and are critical for several functions in a variety of different cell types, including cell differentiation and extracellular stimuli response. MEF2C is expressed in brain and skeletal muscle and its expression is highest during the early stages of postnatal development. It binds specifically to the MEF2 element present in the regulatory regions of many muscle-specific genes. MEF2C is reported to be involved in cardiac morphogenesis and myogenesis, and in vascular development. It is also required for proper development of megakaryocytes and platelets and for bone marrow B-lymphopoiesis. It is shown to be crucial for normal neuronal development, distribution, and electrical activity in the neocortex and plays a role in hippocampal-dependent learning and memory by suppressing the number of excitatory synapses and thus regulating basal and evoked synaptic transmission. Six isoforms of MF2C have been described that are produced by alternative splicing. Isoforms that lack the repressor domain (aa 368-399) are more active. Its β-domain (aa 271-278) that is required for enhancement of transcriptional activity is missing in a number of isoforms. Phosphorylation of MEF2C on serine 59 enhances its DNA binding activity and phosphorylation on serine 396 is required for sumoylation on lysine 391 and inhibition of its transcriptional activity. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
