General description

Metabotropic glutamate receptor 5 or mGluR5 (UniProt P31424; also known as G protein-coupled receptor GRM5, G protein coupled receptor family C, group 1, member E) is encoded by the Grm5 (also known as Gprc1e, Mglur5) gene (Gene ID 24418) in rat species. The metabotropic glutamate receptors belong to a family of G protein-coupled receptors that have been divided into 3 groups based on their sequence homology, putative signal transduction mechanisms, and pharmacologic properties. mGluR1 and mGluR5 constitute Group I metabotropic glutamate receptors. mGluR5 is found on postsynaptic terminals of neurons and microglia. mGluR5 signals via Gq/G11 coupling to activate phospholipase C, resulting in calcium mobilization and activation of protein kinase C (PKC). In addition, mGluR5 is also reported to mediate signaling involving Src-family protein tyrosine kinase in a G protein-independent manner.

Specificity

Expected to react with all spliced isoforms of human, rat, and mouse mGluR5 reported by UniProt (P41594, P31424 and Q3UVX5, respectively).

Immunogen

Epitope: C-terminal cyoplasmic domain.
Synthetic peptide corresponding to a sequence from the C-terminal cytoplasmic domain of rat mGluR5.

Immunocytochemistry Analysis: An 1:100 dilution from a representative lot detected Metabotropic Glutamate Receptor 5 in rat cortical neurons.
The unconjugated antibody (Cat. No. AB5675) is shown to be suitable also for Western blotting and immunohistochemistry applications.
