General description

DVL2 is a member of multigene family that also contains DVL1 and DVL3. The vertebrate dishevelled proteins have approximately 40% amino acid sequence similarity with Drosophila dsh. DVL2 encodes a 90-kD protein that undergoes posttranslational phosphorylation to form a 95-kD cytoplasmic protein.

Specificity

Disheveiled-2 protein (dvl-2). By Western blot the antibody shows no cross reactivity with recombinant disheveiled-1 (dvl-1) or disheveiled-3 (dvl-3) protein. Pre-adsorption of the antibody with the immunogen peptide completely abolishes its reactivity by Western blot against disheveiled-2 protein.

Synthetic peptide corresponding to amino acids 600-614 of human and mouse disheveiled-2 protein.

Western blot: 1:1,000 using ECL. The antibody reacts with a protein of ~95kDa. Faint reactivity was seen in the dvl-1 and dvl-3 HEK293 cell lysates suggesting there may be endogenous dvl-2 present.

Immunohistochemistry: 1:1,000 using the avidin-biotin system.

Dilute antibody in 50 mM PBS, pH 7.4 containing 1.5% sodium chloride and 1% normal animal serum (same species as the host of the secondary antibody).

Optimal working dilutions must be determined by end user.

Partially purified immunoglobulin. Liquid in PBS, pH 7.4, containing 0.01M sodium azide.

Storage and Stability

Maintain at -20°C in undiluted aliquots (not less than 10 μL) for up to 6 months. Avoid repeated freeze/thaw cycles. The use of high quality antiserum-grade plastic or glass is recommended.
