Application

Each vial contains >=1X106 viable cells.

Cells are tested negative for infectious diseases by a Mouse Essential CLEAR Panel and have been found to be negative for all pathologies in this test.

Cells are verified to be of mouse origin and negative for interspecies contamination from rat, human, Chinese hamster, Golden Syrian hamster, and non-human primate (NHP) as assessed by a Contamination Clear panel.

Cells are negative for mycoplasma contamination.

Cells are genotyped by STR analysis to verify the unique identity of the cell line.
Melanoma is a type of skin cancer that develops in melanocytes, the cells responsible for production of pigments. Spontaneous genetic mutations have been associated with some melanomas. Immunotherapies targeting these mutated genes or pathways have been proven to have significant benefits for melanoma patients by improving therapeutic response and survival. The OSUMMER (Ohio State University and Moffitt Melanoma Exposed to Radiation) cell lines satisfy a previously unfilled gap in melanoma biology. These lines are derived from the TN mouse model of melanoma. These mice are homozygous for the LSL-Nras Q61R allele. The oncogene expression is activated by tamoxifen-inducible Cre recombinase in melanocytes. Loss of function is an early event observed in the majority of human melanomas; TN mice additionally carry p16INK4A conditional knockout alleles. The OSUMMER.13 mouse melanoma cell line was derived from a female TN mouse exposed to UVA and UVB radiation. OSUMMER.13 carries the specific NRAS mutation Q61R.WES revealed high SNV (single nucleotide variants) and low CNA (copy number alterations) burden, as well as the presence of UV signature mutations. OSUMMER.5 cells express the melanoma markers PMEL and MelanA, and exhibit robust growth in culture.SourceOSUMMER.13 was derived from melanoma induced in TN (Tyr::CreER) mice exposed to UVA or UVB radiation on postnatal day 3.References1. Features and Benefits

This OSUMMER.13 female mouse NRAS-mutant cell line is a UV-accelerated model for melanoma obtained from the TN model of mouse melanoma. This model is syngeneic with the C57BL/6 mouse model. OSUMMER cells can form tumors in vivo without Matrigel injection.

Storage and Stability

OSUMMER.13 cells should be stored in liquid nitrogen until use. The cells can be cultured for at least 10 passages after initial thawing without significantly affecting the cell marker expression and functionality.
