General description
4-Hydroxyazobenzene-2-carboxylic acid (HABA) is an avidin-binding dye that can be stoichiometrically displaced by biotin. The subsequent change in absorption at A500 provides a basis for spectrophotometric determination of biotin levels in biological samples.
HABA/Avidin Reagent is specific for chromogenic applications that depend upon quantification of biotin.
Application
HABA/Avidin Reagent is suitable for spectrophotometric determination of biotin and biotin conjugates in a wide variety of samples.
Reagent is capable of detecting biotin at μMolar concentrations (i.e., 1 nmole per ml). Typically used to quantitate biotin in biotinylated compounds.
Reconstitution
Prepares a ready-to-use HABA/avidin solution in buffered saline, pH approx. 7.3, with preservative when reconstituted with 10 ml deionized water.

Parameters and conditions of an enzyme based assay for biotin and avidin are presented. Biotinylated glucose-6-phosphate dehydrogenase when complexed with avidin becomes inactivated. Thus it was possible to construct a competitive assay system for biotin. The assay is sensitive between
